In a 6-week two-arm parallel-group randomised double-blind placebo-controlled trial in 100 adults aged 18 to 45 reporting dissatisfied sleep, magnesium L-threonate at 2 g daily produced greater improvement than placebo on the NIH Cognitive Toolbox Total Cognition Composite (p = 0.043), with larger treatment effects on working and episodic memory, greater improvement in reaction time (p = 0.031) and in sleep-related impairment (p = 0.043). There were no group differences on Raven Progressive Matrices (p = 0.953), sleep disturbance (p = 0.316), restorative sleep (p = 0.439), or on any sleep outcome recorded by the wearable tracker. In a randomised double-blind placebo-controlled trial in 83 advanced cancer patients taking oral opioids, it slowed the rise in daily oral morphine dose from day 30 onward.
