In soy the compound circulates almost entirely as glucoside and malonylglucoside conjugates, which are too polar to cross the gut wall efficiently. Bacterial beta-glucosidases cleave the sugar to release the absorbable aglycone. How much a person absorbs therefore depends on which bacteria they carry, which is one reason isoflavone blood levels vary so widely between people on the same intake.
Several Lactobacillus strains express beta-glucosidase and are used commercially to pre-convert soy isoflavone glucosides during fermentation. Co-formulating or fermenting shifts the delivered material toward the aglycone form. Strain matters here, and not every Lactobacillus product carries the relevant enzyme activity.
Fermentable fibre feeds the colonic bacteria that carry out isoflavone deconjugation and downstream metabolism. Shifting the community can shift how much aglycone is liberated. The direction is plausible and the size of the effect in people has not been pinned down.
Isoflavone phenols and ascorbate sit in overlapping redox territory, with ascorbate able to regenerate oxidised phenolic species in vitro. This is chemistry measured in solution and in cell systems. It is a mechanism, not a demonstrated benefit in people.
Soy isoflavone material is routinely combined with calcium and vitamin D in products aimed at bone mineral support. The pairing is conventional rather than mechanistically linked, and no meaningful effect of calcium on isoflavone absorption has been reported. Read it as formulation practice.
MK-7 is produced by Bacillus subtilis natto fermentation of soybeans, so the two constituents already travel together in traditional fermented soy foods. They act through unrelated routes, vitamin K2 through carboxylation of bone matrix proteins. The pairing is compositional, not synergistic in any measured sense.
