Tannins and gallotannins in Java plum seed bind non-heme iron in the gut lumen and cut its uptake. Ascorbate reduces ferric to ferrous iron and forms a soluble complex, which offsets part of that binding. Taking iron with vitamin C and away from the polyphenol dose is the standard way this is handled.
Polyphenols with adjacent hydroxyl groups chelate divalent cations, and zinc is among them. A tannin-rich seed extract taken in the same dose as a zinc supplement can reduce how much zinc is absorbed. Separating the two by a couple of hours is the usual formulation answer.
Java plum seed already contributes hydrolysable tannins including gallic and ellagic acid derivatives. Adding a purified tannin raises the total tannin load, which increases both the mineral binding and the astringency a person notices. Worth flagging rather than combining without thought.
A supplemental amylase is added to help break down starch; Java plum polyphenols inhibit that same enzyme in vitro. Put in one capsule they work against each other. This is the clearest anti-synergy in the ingredient's chemistry.
Tannins precipitate proteins, which is why an astringent extract tastes drying and why polyphenol content of a beverage falls when milk protein is added. In a protein shake the polyphenols bind whey and less free polyphenol remains available. The consequence is loss of measurable free polyphenol, not a safety issue.
Soluble fibre slows gastric emptying and hinders enzyme access to starch by raising luminal viscosity, a different mechanism from polyphenol enzyme inhibition. The two act at separate points on the same process, which is what makes the pairing additive rather than redundant.

Java plum is the fruit of Syzygium cumini. The seed is the part usually extracted for supplements, and it is chemically distinct from the pulp: the seed is dominated by hydrolysable tannins, gallic acid and ellagic acid derivatives, while the pulp and skin carry anthocyanins.
Gallotannins and ellagitannins bind digestive alpha-amylase and alpha-glucosidase non-covalently and reduce their activity in vitro. This enzyme inhibition is the mechanism most often cited for the seed extract's effect on carbohydrate handling.
Polyphenols with catechol and galloyl groups chelate divalent and trivalent metal ions, so a tannin-rich extract taken in the same dose as iron, zinc or copper reduces how much of that mineral is absorbed.
Ellagitannins are not absorbed intact. Gut bacteria hydrolyse them to ellagic acid and convert that to urolithins, so what circulates after a dose depends on an individual's microbiota rather than on the label alone.
