Ananain is not usually isolated on its own. It arrives inside stem bromelain preparations, where several cysteine proteases with overlapping but not identical cleavage preferences act on the same protein substrates. Because the specificities differ, the mixture hydrolyses a wider range of peptide bonds than any single component would. Anyone taking a bromelain product is already taking some ananain, whether or not the label says so.
Papain from papaya latex and ananain from pineapple stem share the same catalytic architecture but cleave at somewhat different residues. Blending them broadens the range of peptide bonds attacked in a mixed protein meal. Both are also inactivated by the same conditions, so the pairing does not extend the pH window or the heat tolerance of either one.
The catalytic residue of ananain is a cysteine thiol. Oxidised to a disulfide or blocked by a heavy metal ion, it stops working. Free cysteine keeps the site reduced, which is why laboratory activity measurements routinely include a thiol reagent. Whether that matters in a finished capsule depends on how much oxygen and how many trace metals the powder has been exposed to, which is a formulation question rather than a physiological one.
Glutathione keeps thiol groups reduced and can maintain the catalytic cysteine of papain-family enzymes in its active form in vitro. This is a stability and activity relationship measured in solution, not a demonstrated effect of swallowing the two together. Read it as mechanistic rather than clinical.
Plant cysteine proteases work over a wide pH range and are usually paired with amylase, lipase and sometimes a fungal or bacterial protease in a single blend. Each enzyme handles a different macronutrient, so the combination is about coverage rather than any interaction between the enzymes themselves. The blend is formulation convention.
Ananain cleaves intact protein into shorter peptides, which is the same chemistry used industrially to make hydrolysed whey. Adding a plant protease to a protein drink pre-digests some of the substrate, and in a healthy gut pancreatic proteases would have done that anyway. The pairing matters more for people with reduced pancreatic output than for someone whose digestion is unremarkable.
The catalytic thiol that makes ananain work is also what makes it vulnerable. Metal ions with affinity for sulfur coordinate to that residue and shut the enzyme down in solution. Taking a high-dose zinc capsule in the same swallow as a protease blend is not a documented clinical problem, but separating them costs nothing if enzyme activity is the point.
Betaine hydrochloride is taken to lower stomach pH. Stem proteases retain some activity across a wide pH range but are progressively denatured in a strongly acidic stomach. Pairing an acidifier with an uncoated protease works against the enzyme, while an enteric coat sidesteps the question by releasing past the stomach.

It is an enzyme squeezed out of pineapple stems, the same source as bromelain, and sold by how much protein it can chop up rather than by weight.
Made from a plant. What ends up in the capsule tracks the harvest, so batch testing and a stated marker matter more here than with a made molecule.
Ananas comosus stems, a by-product of fruit harvesting, are the commercial source. The fruit contains a different protease profile from the stem.
Stems are ground and pressed, and the crude juice is filtered to remove fibre and particulates.
Proteins are concentrated by ammonium sulfate precipitation or ultrafiltration, then separated by ion-exchange or affinity chromatography, which is the step that distinguishes ananain from stem bromelain.
Material is standardised on proteolytic activity units measured against casein or a synthetic substrate rather than on enzyme mass.
The concentrate is dried with a carrier and either filled directly or coated with an acid-resistant polymer.
