Tannin-rich root preparations form insoluble complexes with non-heme iron before it reaches the absorptive surface. The practical consequence is lower iron uptake when a tannin-heavy decoction and an iron dose land in the same window. Separating them by a couple of hours is the usual formulation answer. This is a chemistry-level interaction, not a clinical finding in people taking trillium.
Saponins are amphiphilic and partition into the same micelles that carry plant sterols across the unstirred water layer. When both are present in quantity, each reduces the micellar room available to the other. That matters for how much sterol actually gets absorbed, which is a marker of uptake and not an outcome anyone has measured for trillium specifically.
Dietary saponins can lower the absorption of fat-soluble vitamins by altering micelle stability and by binding bile acids. Direction depends on dose: small amounts behave as surfactants that help solubilise, larger amounts sequester bile salts and work the other way. No human work exists on trillium saponins at either end of that range.
Saponins are surface-active and can provoke nausea or gastric burning at higher doses. Herbal practice has long paired harsh rhizomes with a carminative to make the preparation easier to keep down. This is a tolerability convention rather than an effect anyone has quantified.
The aglycone forms released after gut hydrolysis are poorly water soluble and need a lipid carrier to stay dispersed. A medium-chain triglyceride base keeps them in solution through the stomach and into the duodenum. This is standard formulation reasoning for steroidal sapogenins and has not been tested with trillium extract in people.
