Sunifiram is a piperazine derivative, structurally distinct from the racetams, and no combination study grounds this pairing. It is a convention carried over from a different chemical class.
Caffeine raises arousal through adenosine receptor antagonism, an entirely separate route from glutamatergic signalling. Stacking two arousal raising compounds increases the chance of overstimulation, restlessness and disturbed sleep. This is a caution worth stating, not a recommended combination.
L-theanine is a weak glutamate analogue with mild calming effects and is commonly used to blunt overstimulation from other compounds. Any interaction with a glutamatergic agent is speculative and untested here. No dose relationship has been established for the pair.

Sunifiram, also called DM-235, is a synthetic piperazine derivative developed from the ampakine research line; it is not a nutrient, has no dietary occurrence and no established nutritional role.
The published pharmacology for this compound is rodent work describing enhancement of glutamatergic signalling at AMPA and NMDA receptor sites; rodent receptor pharmacology does not establish a human effect.
Compounds acting on glutamatergic transmission carry a dose dependent excitotoxicity concern in preclinical models, which is why the therapeutic window matters more here than for a nutrient with a wide intake range.
