Pepsin starts protein breakdown in the acid stomach and prefers bonds next to aromatic residues; elastase continues the job downstream at near neutral pH with a different bond preference. The two act in sequence on the same protein, which is why some blends list both. Pepsin needs acid and elastase does not tolerate it, so dose forms usually separate them.

An acute bovine colostrum crossover study tracked neutrophil elastase as a marker of white cell activation during prolonged cycling. That protein is neutrophil elastase, a different enzyme from the pancreatic digestive elastase sold in enzyme blends. What Elastase actually does.

Pancreatic elastase is a serine protease of the chymotrypsin family. It cleaves peptide bonds on the carboxyl side of small uncharged residues such as alanine, glycine and valine, a specificity that complements trypsin and chymotrypsin rather than duplicating them.

The pancreas secretes it as proelastase. Trypsin activates it inside the duodenal lumen, which keeps the protease inert while it is still inside pancreatic tissue and ducts.

Its working pH range sits near neutral, matching the bicarbonate-buffered duodenum. Gastric acid and pepsin denature the free enzyme, which is why oral pancreatic enzyme products are either enteric coated or dosed with a meal that buffers the stomach.

Faecal elastase-1 resists breakdown during intestinal transit, so its concentration in stool is used as a marker of how much enzyme the pancreas secreted. A stool reading reports secretory output, not how well a meal was digested.

Where Elastase comes from.

Almost all of it comes from pig or cow pancreas, collected at meat processing and purified into the enzyme mixture called pancreatin. A different route grows proteases in a fermentation tank for vegetarian products. Strength is measured by what the enzyme does, not by how much powder is in the capsule, which is why labels list units.

The same molecule is reached more than one way. Which route a given product used is a manufacturing choice, and the finished compound is the same either way.
