What German Knotweed is, and what it does.

Does it work
It suits people who already want resveratrol and prefer the root extract it is actually made from. If a label lists standardised resveratrol, this is very often its origin.
How much to take
No daily amount is on record for the root itself. Standardised extracts are dosed by their resveratrol content, so the stilbene figure on the label is the number to read.
Time to feel it
Nobody has measured a time course for this material in people, so there is no honest duration to quote.
The first dose
Day one is mostly pharmacokinetics. Resveratrol absorbs, then gets conjugated quickly, and nothing measurable shifts on a panel that fast.
With regular use
Weeks of daily use haven't been tracked for the whole root. The stilbene literature that exists mostly follows purified resveratrol rather than the root extract.
How well tolerated
Some knotweed species carry anthraquinones such as emodin, which act on colonic motility, so looser stools are possible. Tannins bind non-heme iron, so space it from iron.
How it feels
Most people describe nothing distinct. Where it does make itself known it is usually digestive, and a decoction of the root tastes astringent.
The overlooked benefit
Most resveratrol on the market never came near a grape. It is extracted from knotweed root, which concentrates stilbenes far more heavily than grape tissue does.

German Knotweed + ResveratrolKnotweed species in the Polygonum and Reynoutria group are the main commercial botanical source of resveratrol and its glucoside piceid.

Most resveratrol on the supplement market is extracted from knotweed root rather than from grapes, because the root concentration is far higher. Piceid, the glucoside form, has to be deglycosylated before the aglycone is absorbed. Anyone stacking a knotweed extract with a resveratrol isolate is largely doubling the same molecule.

German Knotweed + PterostilbeneStructural analogue of resveratrol with two methoxy groups in place of hydroxyls.

Pterostilbene resists phase II conjugation better than resveratrol does, so it survives first pass more completely. The two hit overlapping targets, meaning the combination is not additive in the way two unrelated ingredients would be. Stacking is common practice with little human comparison behind it.

German Knotweed + QuercetinBoth are substrates for the same sulfotransferase and UDP-glucuronosyltransferase enzymes in gut wall and liver.

Quercetin competes for the conjugating enzymes that clear stilbenes rapidly, which is the standard rationale for pairing them. The competition is documented in enzyme and cell systems. Whether it produces a meaningful change in circulating stilbene levels in people is not settled.

German Knotweed + IronPolygonum species are tannin-rich, and tannins bind non-heme iron in the gut lumen.

Knotgrass and its relatives carry appreciable condensed and hydrolysable tannins, which form insoluble complexes with dietary and supplemental iron. Taking a knotweed preparation alongside iron works against the iron. Separating them across the day is the practical answer.

German Knotweed + Vitamin CAscorbate reduces polyphenol oxidation in solution and partially offsets tannin-driven inhibition of iron uptake.

Ascorbate keeps iron in the ferrous form and competes with tannins for it, blunting but not removing the inhibition. It also slows the browning of polyphenol extracts during storage. This is class chemistry, applicable to any tannin-rich botanical.

German Knotweed + Milk thistle (silymarin)Both are flavonoid and polyphenol preparations processed through the same conjugating enzyme systems.

Silymarin and stilbenes are both heavily glucuronidated on first pass, so co-dosing means competition for the same limited enzyme capacity. That can nudge exposure of either one up. It is a pharmacokinetic observation from the class rather than a tested pairing.
