What Lanolin is, and what it does.

Does it work
Suits anyone using a barrier balm on dry, chapped skin, and anyone taking vitamin D3, which is most often made from it. It is an animal derived material.
How much to take
No dose figure is on record for lanolin itself. Topically it is a thin layer where skin is dry. In capsules it is the source material for vitamin D3, not an amount taken.
Time to feel it
On skin the sealing effect is there within minutes to hours. As a vitamin D3 source, the change lands on a blood panel over weeks.
The first dose
Skin feels coated and softer within the hour. As a D3 source, day one is quiet and the change registers on a blood test later rather than as a sensation.
With regular use
Used regularly on dry areas it keeps water loss through the skin down and skin stays more supple. As a D3 source, weeks of intake move vitamin D status upward.
How well tolerated
Well tolerated by most, though contact sensitivity does happen, mostly to the free lanolin alcohol fraction. Purified, reduced alcohol grades exist for reactive skin.
How it feels
Thick, waxy and slow to sink in. Skin feels sealed rather than watered, because the film holds moisture in rather than adding any.
The overlooked benefit
It absorbs about twice its own weight in water. That is how hydrous wool fat works, a wax that carries water into a cream without an added emulsifier.

Lanolin + Vitamin D3Lanolin is the industry feedstock for cholecalciferol: 7-dehydrocholesterol is isolated from lanolin and converted by ultraviolet irradiation.

Almost every non-vegan vitamin D3 on the market starts as sheep wool wax. The 7-dehydrocholesterol fraction is separated from lanolin, irradiated with ultraviolet light to open the B ring, and thermally isomerised to cholecalciferol. The finished D3 contains no lanolin, which is why it clears a wool allergy concern but not a vegan one. This is the single most important relationship lanolin has in supplements.

Lanolin + MCT oilCholecalciferol is fat soluble and its absorption depends on being presented in a lipid phase.

Lanolin-derived D3 is a crystalline solid with almost no water solubility. Dissolving it in a medium-chain triglyceride carrier puts it into the mixed micelle phase the small intestine actually absorbs from. Softgels use this for a reason, and dry tablets taken without any fat are the weaker case. The oil is the vehicle, not an active.

Lanolin + Vitamin K2 MK7Both are fat-soluble vitamins routinely co-formulated, and both act on calcium handling at different points.

Lanolin-derived D3 raises calcium absorption from the gut, while K2 is the cofactor for carboxylating the matrix Gla protein and osteocalcin that direct where calcium is deposited. Formulators pair them because the two steps sit at opposite ends of the same handling chain. They share the same lipid vehicle, which makes the combination straightforward to make. The pairing is formulation convention backed by clear biochemistry.

Lanolin + MagnesiumThe hydroxylase enzymes that convert cholecalciferol to 25-hydroxyvitamin D and then to the active hormone are magnesium dependent.

Cholecalciferol from lanolin is inert until two hydroxylation steps run, one in the liver and one in the kidney. Both enzymes require magnesium, as does the vitamin D binding protein interaction. Low magnesium status means a dose gets converted less efficiently. This is textbook cofactor biochemistry, not a marketing pairing.

Lanolin + CalciumThe active vitamin D metabolite drives intestinal calcium absorption through calbindin and TRPV6 expression.

Lanolin-sourced D3 is what makes dietary calcium usable, by upregulating the transport machinery in the duodenum. Calcium taken without adequate vitamin D status is absorbed at a much lower fraction. The two are co-dosed for exactly this reason. Combining them says nothing about bone density on its own.

Lanolin + Hyaluronic acidTopical formulation logic: an occlusive and a humectant do different jobs in the same product.

Lanolin works as a semi-occlusive layer that slows water loss from the skin surface. Hyaluronic acid works from the other direction by holding water in the upper layers. A humectant applied without something above it can dry out in low humidity, which is where the occlusive matters. This is skin formulation practice, not systemic biology.

Lanolin + CeramidesBoth contribute to the lipid component of the skin barrier, from outside and within the lamellar structure respectively.

Lanolin's sterol esters and branched fatty acids sit on the surface and reduce transepidermal water loss. Ceramides are structural lipids of the stratum corneum itself. Products combine them to address the barrier at two levels. Evidence for the combination specifically is thinner than evidence for either alone.

Lanolin + SqualaneBoth are emollient lipids used to adjust the feel and spreadability of an occlusive base.

Neat lanolin is tacky and heavy, which limits where people will use it. Squalane is light and spreads easily, so blending it cuts the drag without removing the occlusive effect. This is a texture decision made by the formulator. Neither ingredient changes the other's chemistry.

Lanolin + Vitamin ELanolin contains unsaturated and branched fatty acid esters that oxidise on storage, and tocopherol is the standard antioxidant used to slow that.

Oxidised lanolin is more irritating than fresh lanolin, and the free lanolin alcohols released by oxidation are the fraction most often linked to contact sensitivity. Adding tocopherol to the fat phase slows peroxide formation during shelf life. This protects tolerability rather than adding any activity. It is why grade and freshness matter more than concentration.

Where Lanolin comes from.

Lanolin is sheep wool grease, collected when fleece is washed. It ends up in two places: thick barrier creams, and vitamin D3, because the cholesterol-like molecule inside it turns into vitamin D under ultraviolet light exactly the way it does in your skin.
