Licorice is the conventional partner for harsh bitter roots, added for palatability and to soften gastric irritation. Glycyrrhizin is also a surfactant glycoside that improves the dispersion of poorly water-soluble constituents. The pairing carries licorice's own sodium and potassium handling caution, which does not disappear because the dose is small.
Ginger is added to bitter cold formulas to counter the nausea and gastric discomfort those formulas can cause. The reasoning is empirical practice rather than a measured interaction. Nothing about the pairing changes the alkaloid load being taken.
Berberine and the sophora alkaloids are both bitter, poorly absorbed plant bases with substantial microbial handling in the gut. Taken together the gastrointestinal burden adds up, and both undergo microbiota-dependent conversion before absorption. Read this as a caution on stacking bitter alkaloids, not as a productive combination.
Oxymatrine is converted to matrine by intestinal bacteria, so the composition of the gut community sits directly on the absorption path. Anything that shifts that community can shift how much matrine actually appears. This is a real mechanistic dependency and not a reason to expect a probiotic to increase the effect in any measured way.
Quinolizidine alkaloids have documented central effects at the upper end of intake, including drowsiness and, at toxic doses, worse. Combining them with sedating botanicals stacks that direction without any way to measure the total. This is flagged as an anti-synergy, meaning the pairing warrants avoidance rather than encouragement.
Matrine is cleared hepatically, and silymarin is the conventional botanical added to formulas where hepatic load is the concern. No combination study exists. Adding silymarin is a formulation habit and should not be read as licensing a higher sophora dose.

Two of this root's alkaloids can convert into each other, and gut bacteria turn one form into the other before absorption, so taking one of them acts in part like a delayed dose of the other.
A couple of its flavonoids have an extra chemical group that makes them much more fat-soluble than typical flavonoids, so they tend to settle into cell membranes and aren't well extracted by water alone.
Because the alkaloids in this root are chemically basic, how much you extract depends heavily on the acidity of the extraction process, so plain water, acidified water and alcohol extracts all end up with different alkaloid amounts, meaning an extract ratio alone doesn't tell you what's actually in it.
