Vitex negundo carries flavonoids such as vitexin and casticin plus iridoid glycosides, and those classes are heavily glucuronidated during first pass. Vitex agnus-castus and Vitex negundo share iridoid glycosides such as agnuside and a similar flavonoid backbone. Work on Vitex negundo in a rodent model of ovarian dysfunction reported changes in endocrine and inflammatory markers, which is where the overlap is claimed. Stacking two Vitex species means stacking the same constituent classes, so the pairing raises total exposure rather than adding a distinct action. The leaf and seed contain flavone compounds including casticin, iridoid glycosides, and lignans, with casticin used as the main marker for standardizing extracts.
These flavones don't dissolve well in water and get heavily modified by the gut and liver before reaching the bloodstream, so the amount of the original unmodified compound that circulates after taking it by mouth is low.
The iridoid glycosides need gut bacteria to strip off their sugar before the remaining part can be absorbed, so how much gets through depends on each person's gut microbes and varies between individuals.
