Dipeptidyl peptidase IV is a serine exopeptidase that lifts dipeptides off the free front end of a peptide when proline or alanine sits in the second position. That one specificity rule is the whole reason it gets used, because most peptidases can't cut next to proline.

Proline's ring stiffens the peptide backbone, so proline-rich stretches survive ordinary stomach and pancreatic proteases and land in the small intestine as intact fragments. Wheat gliadin peptides are among the most proline-rich sequences in the diet, which is why a proline-specific peptidase gets paired with them.
