The interaction is established for piperine as a class effect, not measured for this specific lignan. The same mechanism can raise exposure to medicines, which is the reason it deserves flagging rather than celebrating.
3,4-divanillyltetrahydrofuran is a lipophilic lignan with poor water solubility, so it depends on bile and dietary fat for uptake. Phospholipid carriers give it a dispersible vehicle. This is standard formulation practice for lipophilic botanicals. It changes delivery, not what the molecule does.
Medium-chain triglycerides give a liquid lipid phase in which a poorly water-soluble lignan can be dissolved and delivered. Softgel products use this routinely. The consequence is more consistent absorption than a dry powder, not a different mechanism of action. Taking the extract with a meal containing fat does the same job.
Calcitriol signalling through the vitamin D receptor is active in the tissues these formulas address, and vitamin D status is frequently low. That makes it a sensible companion rather than a demonstrated partner. No combination work exists. The pairing rests on separate established roles.
DIM is used to move the balance of oestrogen metabolites through phase I hydroxylation. Nettle root lignans act, if at all, on the transport protein side rather than on metabolism. Combining them targets two different steps in the same axis, which is the formulation logic. Neither has been studied with the other.
Quercetin has its own small human literature in urinary comfort and is often placed alongside nettle root in the same product. The two act on different targets. There is no combination trial. Read the pairing as commercial convention with plausible non-overlap.

3,4-divanillyltetrahydrofuran is a lignan built on a tetrahydrofuran ring carrying two vanillyl groups, and it is one of several lignans found in the root of Urtica dioica rather than in the leaf. Leaf and root preparations are chemically different materials.
The molecule is lipophilic and poorly water soluble, so its uptake depends on bile salt micelle formation and on the presence of dietary fat, which is why it is supplied in oil-based or phospholipid-carrier formats.
Nettle root also carries sterols such as beta-sitosterol, polysaccharides and a lectin, so a whole-root extract standardised on 3,4-divanillyltetrahydrofuran still delivers several other constituent classes alongside it.
Because 3,4-divanillyltetrahydrofuran occurs in root tissue at low natural abundance, standardised percentages on labels typically refer to a small fraction of the extract mass, and a synthesised isolate is a different material from a standardised root extract.
Where Nettle Root (3,4-DVT) comes from.
There are two ways to get it. One is to dry nettle roots, soak them in an alcohol and water mix, and concentrate what comes out; that also brings along the other things in the root. The other is to build the molecule in a lab, which gives you the pure compound and nothing else. Labels say which by naming either a root extract or the isolated compound.
The same molecule is reached more than one way. Which route a given product used is a manufacturing choice, and the finished compound is the same either way.
Stinging nettle roots and rhizomes, harvested from cultivated or wild stands, then washed and dried. Root and leaf are different raw materials and are not interchangeable.
Milled root is extracted with ethanol and water; ethanol content determines how much of the lipophilic lignan fraction is carried over versus the water-soluble polysaccharides.
The solvent is stripped under reduced pressure and the concentrate is dried, usually by spray or vacuum drying, onto a carrier.
Some products supply 3,4-divanillyltetrahydrofuran made by chemical synthesis from vanillin-type building blocks rather than isolated from root, which yields the same molecule with none of the plant matrix.
