The pairing appears in relaxation-positioned formulas. The mechanistic link is real; a combination outcome has not been measured here.
Theanine is a glutamate analogue that shifts cortical activity toward alpha rhythm, a different route from the monoamine pathway 5-HTP feeds. Formulas combine them for evening use on that complementary basis. Anything additive on alertness is worth stating plainly rather than assuming it is only beneficial.
Glycine acts at its own inhibitory receptor and at the NMDA receptor glycine site, unrelated to serotonin synthesis. It is paired with 5-HTP in evening formulas for that separate inhibitory action. The combination is formulation convention, not a tested pair.
GABA is the principal inhibitory neurotransmitter and is often stacked with 5-HTP in relaxation formulas. Caffeine antagonises adenosine receptors and pushes alertness in the opposite direction from an evening-positioned serotonin precursor. Taking them together works against the reason each is in the formula. This is a timing conflict worth stating, not a chemical incompatibility.
Tryptophan hydroxylase, the rate-limiting enzyme that makes 5-hydroxytryptophan from tryptophan, is an iron-dependent monooxygenase. Supplemental 5-HTP enters the pathway after that step, so it does not depend on iron status the way endogenous synthesis does. The relationship is upstream and explains where 5-HTP sits in the sequence.
Several serotonin receptor subtypes signal through phospholipase C and the phosphatidylinositol second messenger system, for which inositol is the substrate. That makes inositol a downstream signalling component rather than a synthesis partner. The link is mechanistic and has not been tested as a combination.

The enzyme that turns 5-HTP into serotonin needs the active form of vitamin B6 as its helper, which is why your B6 status sits directly on this pathway.
Tryptophan has to queue with other large amino acids for the same doorway into the brain. 5-HTP does not use that queue, so a meal with protein in it does not hold it up the same way.
The converting enzyme also lives in tissues outside the brain, so a share of an oral dose gets turned into serotonin out in the body before it ever reaches the central nervous system.
