What Black Mustard is, and what it does.
- Does it work
- Suits cooks and anyone interested in isothiocyanate chemistry from a food. People whose iodine intake is already thin will want to know about the thiocyanate interaction.
- How much to take
- No daily amount is on record. Start at culinary amounts, and note that an enzyme active powder and a heat treated one give very different isothiocyanate yields.
- Time to feel it
- The pungency is immediate, within seconds of crushing or chewing. Urinary markers of isothiocyanate exposure appear within hours of the meal.
- The first dose
- Day one is heat on the tongue and in the nose. The rest happens in metabolism, where isothiocyanates are joined to glutathione and cleared.
- With regular use
- Weeks of regular brassica intake keep isothiocyanate exposure steady, and that is measurable in urine. It's an exposure marker, not an outcome.

- In food amounts it's an everyday spice. The concentrated oil burns skin and mucous membranes. If your iodine intake is low or you take thyroid medicine, ask your doctor first.
- How it feels
- Sharp, nose clearing heat that hits the sinuses more than the tongue and fades fast. On skin it feels warm first and then stinging.

- Heat kills myrosinase, so a cooked powder can be full of glucosinolate and still yield almost no isothiocyanate. Content and yield are two different measurements.

Why these belong in the same formula. Each row says what the basis is, from settled biochemistry through to a trial that measured the pair.
Glucoraphanin is a storage form and does nothing until myrosinase cleaves it to sulforaphane. Mustard seed powder carries a myrosinase that can perform that cleavage in the gut when the broccoli material no longer can. A 2026 human study measured higher sulforaphane bioavailability from a glucoraphanin-rich broccoli preparation when exogenous mustard-seed myrosinase was added. This is a conversion effect, not an additive dose effect.
Sprout extracts are standardised on glucoraphanin content, not on sulforaphane, and many have lost myrosinase activity to processing. Adding mustard seed supplies the missing enzyme step. Without it, conversion depends on gut bacteria, which varies widely between people. The pairing is a formulation answer to that variability.
Myrosinase activity is stimulated by ascorbate at low millimolar concentrations, a long-established feature of the enzyme's biochemistry. In a formulation pairing mustard-derived myrosinase with a glucosinolate source, vitamin C is a plausible cofactor addition. The effect is on the enzyme step rather than on absorption of the end product. It has been characterised in plant and in vitro systems rather than in a human supplement trial.
Thiocyanate and related metabolites are competitive inhibitors of iodide uptake into thyroid tissue. This matters mainly where iodine intake is already low. With adequate iodine the competition is of little consequence. Anyone taking large amounts of brassica seed material should keep iodine intake adequate rather than avoid one or the other. The mechanism is settled pharmacology.
Isothiocyanates released from mustard glucosinolates upregulate Nrf2-driven phase II enzymes. Some downstream antioxidant enzymes, notably the glutathione peroxidases, require selenium as selenocysteine. The overlap is mechanistic and plausible rather than demonstrated in a human co-supplementation trial. Read it as pathway complementarity, not as a measured additive outcome.
Every absorbed isothiocyanate is handled by conjugation to glutathione, which is what produces the urinary dithiocarbamate metabolites used to measure exposure. Higher isothiocyanate load therefore draws on glutathione turnover. Whether supplemental glutathione changes that flux in people is not established. The conjugation step itself is textbook biochemistry.
Cysteine availability is the rate-limiting input to glutathione synthesis, and glutathione is what conjugates absorbed isothiocyanates. On paper NAC supports the disposal pathway. There is no human trial pairing the two, so this stays mechanistic. Faster conjugation is also not automatically a better outcome, since some isothiocyanate activity depends on intracellular residence.

Black mustard seed keeps a compound called sinigrin separate from the enzyme that acts on it. Crushing or chewing the seed brings them together, and that reaction produces the pungent compound responsible for mustard's bite.
That enzyme breaks down with heat. Cooked or heavily processed brassica material loses most of that enzyme activity, which is why the raw compound content and the amount of active product it actually yields are two different things to measure.
Once absorbed, these pungent compounds get tagged and cleared out through urine as a specific breakdown product. Measuring that breakdown product in urine is the standard way to track exposure, and a marker of exposure is not the same as a marker of effect.
A breakdown product of this compound competes with iodide for uptake into cells. This matters more, and scales with dose, when someone's iodine intake is already borderline.
Where Black Mustard comes from.
It is the seed of the black mustard plant. Crush it and two things that were kept apart inside the seed meet and react, which is where the heat comes from.
Made from a plant. What ends up in the capsule tracks the harvest, so batch testing and a stated marker matter more here than with a made molecule.
Small dark seeds of black mustard, grown mainly in South Asia, Eastern Europe and the Mediterranean. Sinigrin content varies with cultivar, soil sulphur and growing conditions.
Seed is cleaned, sometimes dehulled, then either cold-pressed to remove fixed oil or used whole. The press cake retains most of the glucosinolate.
Milling is the step that decides everything downstream. Low-temperature milling keeps myrosinase active. Hot milling or subsequent heat treatment inactivates it while leaving sinigrin in place.
Material is assayed either for sinigrin content or, for enzyme-active grades, for myrosinase units. These are different specifications and a certificate showing one says nothing about the other.
Ends as a stabilised powder for capsules and tablets, or as steam-distilled allyl isothiocyanate for topical and antimicrobial use.
