Capsaicin activates TRPV1 and raises catecholamine output, while caffeine blocks adenosine receptors and slows cyclic AMP breakdown downstream of those same catecholamines. The two raise metabolic rate through connected steps of one signalling chain.
EGCG slows catechol-O-methyltransferase, the enzyme that clears noradrenaline, while capsaicin raises noradrenaline release through TRPV1 activation. One increases the signal and the other extends how long it lasts.
The 6-paradol and shogaol compounds in grains of paradise activate TRPV1 and related channels, the same family capsaicin acts on, and both raise brown adipose tissue activity. The thermogenic effect is additive on one receptor system.
Gingerols and shogaols are vanilloid compounds that bind TRPV1 much as capsaicin does, adding to the same warming and gastric motility signal. Traditional carminative blends have used the two together for a long time.
Piperine is itself a TRPV1 agonist and also slows intestinal glucuronidation and P-glycoprotein efflux, raising how much of a co-dosed compound survives first pass. It adds pungent signalling and an absorption effect at once.
Salicin metabolites reduce the protective prostaglandin production of the gastric lining, and concentrated capsaicin is a direct mucosal irritant at higher doses. Combining them raises stomach discomfort in sensitive people without adding a shared benefit.
Capsaicin is lipophilic and barely soluble in water, so it partitions into fat and is absorbed passively from the small intestine. Dosing it with a lipid gives it a vehicle instead of leaving it to disperse in gastric fluid. This is a delivery relationship and adds nothing biological of its own.
A phospholipid emulsifier disperses capsaicinoid oleoresin into fine droplets, which raises the surface area available for absorption and evens out how it contacts the gut wall. Emulsified delivery also tends to reduce the concentrated burning sensation a raw oleoresin can produce. The mechanism is formulation chemistry.
Capsaicin activates TRPV1 on sensory nerve endings, and low gastric pH lowers the threshold at which those channels fire. Buffering gastric acid is a plausible way to reduce the upper gut burning that limits tolerance for some people. This is mechanistic reasoning and has not been measured as a pair here.
A gastric acidifier lowers stomach pH, and acid sensitises the same TRPV1 channels capsaicin activates. Taking both together makes upper gut burning more likely rather than less. Separating the doses is the practical response, and this is flagged as a caution not a benefit.
Curcumin is described as a TRPV1 agonist as well, so the two act at least partly on the same sensory channel. Both are also poorly water soluble and depend on the same lipid or emulsified delivery to be absorbed at all. Formulators pair them for the delivery reason as much as the receptor one.
Carnitine carries long-chain fatty acids across the mitochondrial membrane, while capsaicin acts upstream through sympathetic signalling that mobilises fat. The two sit at different points of the same overall process. Combination data is not available, so this stays early.
Glucomannan works physically, forming a viscous gel that slows gastric emptying, whereas capsaicinoid effects on appetite ratings run through sensory and sympathetic signalling. Different mechanisms, so an additive direction is reasonable to expect. Neither the pair nor the magnitude has been measured in the material available here.
