Glucoraphanin is inert until myrosinase cleaves it. Cooking destroys the myrosinase native to broccoli, leaving the glucosinolate dependent on slow and variable conversion by gut bacteria. Raw ground mustard seed carries its own heat-stable-enough myrosinase and can perform that cleavage in the gut. A human crossover study found that adding mustard seed to cooked broccoli raised sulforaphane appearance in the body.

Supplements sold as sulforaphane are frequently glucoraphanin plus a myrosinase source, because glucoraphanin alone converts poorly. Ground mustard seed is one of the practical myrosinase sources. The variable to watch is whether the mustard component has been heat-processed, since heat destroys the enzyme and removes the whole point of including it.

Thiocyanate and iodide are transported by the same symporter, so a high thiocyanate load from brassica glucosinolates reduces iodide uptake. This matters at habitually high brassica intakes combined with low iodine intake, and is why iodine adequacy is the standard consideration alongside heavy mustard or brassica use. Adequate iodine intake removes most of the concern.

When mustard is cooked and its own myrosinase is gone, the remaining glucosinolates travel to the colon where bacterial thioglucosidase activity can cleave them. Conversion by this route is slower and far more variable between people than the plant enzyme route. Which species matter is not settled, so this is a directional mechanism rather than a dosing instruction.

Allyl isothiocyanate and sulforaphane are handled by glutathione S-transferase conjugation, and the resulting mercapturates are what appear in urine. N-acetylcysteine feeds cysteine into glutathione synthesis. The interaction runs both ways: cysteine supply supports the conjugation route, and heavy isothiocyanate intake draws on that same pool. This is metabolic bookkeeping, not a demonstrated clinical benefit of the pairing.

The mercapturic acid pathway conjugates isothiocyanates to glutathione before excretion. This is settled biochemistry and is how isothiocyanate exposure is measured in urine. Whether oral glutathione meaningfully raises the tissue pool is a separate and unsettled question.

Isothiocyanates from mustard glucosinolates activate Nrf2 signalling and raise expression of phase II and antioxidant enzymes. Several of those defensive enzymes are selenium dependent. The pairing therefore makes mechanistic sense, but the combination has not been tested in people at supplement doses.

Mustard seed keeps a compound called sinigrin separate from the enzyme that activates it. Crushing or chewing brings them together, releasing the pungent compound that gives mustard its heat.

That activating enzyme is destroyed by heat, so cooked mustard still has the starting compound but loses most of the enzyme, meaning gut bacteria have to finish the job, which makes the outcome slower and more variable person to person.

The pungent mustard compounds get bound to a natural antioxidant in the body and excreted in urine as specific breakdown products, and that's the standard way researchers measure how much of these compounds someone has actually taken in.
