Selank is a short peptide, and peptide bonds are exactly what pepsin cleaves at gastric pH. Anything that increases gastric proteolytic activity while the peptide is in the stomach reduces the amount that survives intact. This is why the molecule is handled as a parenteral or intranasal peptide rather than an oral one.
Betaine hydrochloride is taken to lower gastric pH, which both activates more pepsin and speeds acid hydrolysis of peptide bonds. For an orally swallowed peptide that works against intact delivery. The interaction disappears entirely for a non-oral route.
Protease-containing enzyme blends are formulated to break proteins and peptides into amino acids and dipeptides. A heptapeptide is a substrate for them, not an exception. Co-dosing an oral peptide with a protease blend reduces how much reaches absorption intact.
Bromelain is a broad-specificity protease active across a range of pH values, including in the small intestine. It cleaves peptide bonds without regard to the peptide's intended purpose. Separate the two if an oral peptide is being used at all.
Papain hydrolyses peptide bonds with wide specificity and stays active over a broad pH range. A seven-residue peptide is well within its substrate range. The practical consequence is loss of intact peptide when the two are taken together orally.
L-theanine has its own human literature for calm attention, and Selank is used with a similar intent. Nothing measured the two together, so this is overlap of purpose rather than a demonstrated additive effect. Anyone stacking calming agents should expect the drowsiness of each to add.
Glycine acts at inhibitory glycine receptors and is used for sleep onset support. Combined with any calming agent the sedative effects can add. There is no combination evidence for this specific pair, and the grounding is glycine's own neurochemistry.
Supplemental GABA acts largely at peripheral receptors because it crosses the blood-brain barrier poorly, so the pairing is weaker than it looks on a label. Where any calming effect does occur, it can add to that of another calming agent. No study of the combination was located.
Magnesium sits in the NMDA receptor channel and contributes to normal control of neuronal excitability, which is why it appears in calming formulas. Additive drowsiness with another calming agent is the practical consideration. This pair has not been measured together.
Caffeine blocks adenosine receptors and increases arousal, which pulls in the opposite direction from a calming agent. Taken together each can mask the subjective signal of the other rather than cancel it. Nothing measured the pair, so this is pharmacology, not a result.
Tryptophan is the dietary precursor for serotonin synthesis by way of 5-hydroxytryptophan. Any agent with reported effects on serotonergic tone is therefore often stacked with it. The overlap is at the level of pathway, not of any measured combination outcome.
