Geniposide is an iridoid glucoside, the principal marker constituent of gardenia fruit (Gardenia jasminoides), built from the iridoid aglycone genipin joined to a glucose unit.
The glucose unit makes the molecule polar and poorly permeable, so oral geniposide is absorbed only modestly in its intact form.
Colonic bacterial beta-glucosidase removes the sugar to release genipin, and this microbial activation step largely determines how much active aglycone the body sees.
