Glycyrrhizin, the principal saponin of Glycyrrhiza uralensis, is a glycoside that mammalian enzymes do not cleave well. Colonic bacteria hydrolyse it with beta-glucuronidase to glycyrrhetinic acid, and that aglycone is the form that reaches the circulation.
Glycyrrhetinic acid inhibits 11-beta-hydroxysteroid dehydrogenase type 2 in the kidney. That enzyme normally converts cortisol to inactive cortisone at the mineralocorticoid receptor, so inhibiting it lets cortisol act there, favouring sodium and water retention and increased potassium excretion.
Because the potassium-losing effect is dose and duration dependent, intake of the whole root and of glycyrrhizin-bearing extracts is conventionally bounded, and formulations that remove the saponin do not carry the effect.
Glycyrrhizin is intensely sweet on the tongue by weight compared with sucrose, with a slow onset and a long lingering finish, which is why the root doubles as a flavour ingredient in herbal preparations.

It is a root, dug up and dried. Makers steep it in water or alcohol, boil the liquid down and check how much of the sweet compound is left. The DGL version has that compound taken back out.
