Quercetin is the aglycone released when rutin is cleaved by gut microbes, so a formula pairing Ruscus with quercetin covers the same flavonoid space by a different route. Quercetin also inhibits several drug-metabolising enzymes, which is worth noting in a stacked formula. The pairing is mechanistic.
Ginkgo flavone glycosides and terpene lactones are associated with peripheral blood flow, while ruscogenins act on venous tone. The two sit on opposite sides of the circulation, arterial inflow versus venous return. Ginkgo also carries an antiplatelet consideration that a combined formula should account for.
Alpha-tocopherol works inside membranes while the flavonoids typically paired with butcher's broom work in the aqueous phase, and ascorbate regenerates tocopheroxyl radicals at the interface. The three cover different compartments. At higher intakes vitamin E carries its own platelet consideration.

The active fraction of butcher's broom rhizome is a group of steroidal saponins, chiefly ruscogenin and neoruscogenin and their glycosides ruscin and ruscoside.
Saponins in the rhizome reduce surface tension and interact with membrane cholesterol, which is the general chemical basis for the gastric irritation that plant saponins can cause at higher intakes.
Butcher's broom is a member of the Asparagaceae and the part used is the rhizome and root rather than the aerial cladodes, which carry less of the saponin fraction.
Standardisation is expressed as percent total ruscogenins measured after acid hydrolysis of the glycosides, so a stated ruscogenin figure refers to the aglycones released rather than the intact saponins present.

The root of a small Mediterranean shrub is dried, soaked in an alcohol and water mix to pull out the active saponins, then concentrated and dried onto a carrier so each batch carries the same declared amount.
