- Start with 500 to 2,000mg a day of oil with a declared GLA percentage, taken with food, and keep it running across cycles. The 4,000mg used in trials is a research condition.

- Two to three cycles. This one is measured in months rather than days, because membrane fatty acids turn over slowly.

- Only about seven to ten percent of the oil is GLA and the rest is largely linoleic acid, so a stated GLA percentage tells you more than the capsule size does.

Zinc status sets delta-6 desaturase activity, the enzyme that converts linoleic acid to GLA. Adequate zinc keeps the endogenous route working alongside the supplied GLA.
Ascorbate regenerates spent tocopherol back to its active form. The pair keeps the polyunsaturated oil from peroxidising.
GLA feeds prostaglandin E1, which lowers normal platelet aggregation, and ginkgolides block platelet activating factor. The two effects add on the same physiology.

Gamma-linolenic acid is the compound evening primrose oil is taken for, present at roughly seven to ten percent of the oil. Adding isolated GLA to the same formula raises the same active, so the two are one dose and not two. Any total GLA figure should be read across both.
Delta-6 desaturase, the rate-limiting enzyme that converts linoleic acid to GLA, depends on adequate pyridoxine, zinc and magnesium status. Evening primrose oil supplies GLA already made, so it bypasses that enzyme, but the next elongation and desaturation steps still sit in the same nutrient-dependent pathway. B6 is also long-standing formulation practice in cyclical comfort products.
Magnesium is required by the desaturase and elongase steps that handle GLA downstream, and it is one of the standard companions in formulas aimed at comfort across the menstrual cycle. The cofactor relationship is settled biochemistry; the cyclical comfort use rests on magnesium's own literature, not on the oil's.
GLA elongates to dihomo-gamma-linolenic acid, and DGLA can then be desaturated onward to arachidonic acid by delta-5 desaturase. EPA restrains that delta-5 step, so co-dosing tends to hold more of the GLA at the DGLA stage. This is why the two are often deliberately combined rather than kept apart, and it is a pathway argument rather than an outcome trial.
Linoleic acid is the substrate that feeds the same delta-6 desaturase and elongase machinery GLA uses, and it is by far the larger part of evening primrose oil itself. A large separate linoleic load competes for the same enzymes. In practice this means the GLA percentage, not the total oil volume, is the number that matters.
Alpha-linolenic acid from flax competes with linoleic acid and GLA for delta-6 desaturase and the shared elongases. Combining the two oils splits enzyme capacity between the omega-6 and omega-3 branches. This is a well-described competition in fatty acid metabolism and not a reason to avoid either.
Borage seed oil carries GLA at roughly twice to three times the share evening primrose oil does. Running both means one active from two sources, so the combined GLA figure is what should be counted. The oils differ in their other fatty acids and in their minor constituents.
DGLA-derived eicosanoids tend to dampen platelet aggregation, and nattokinase acts on fibrin and clot handling. The directions add up, which is a caution to flag for anyone on anticoagulant or antiplatelet medication or facing surgery, not a benefit to sell. No combination trial quantifies the overlap.
Willow bark supplies salicylates, which inhibit platelet cyclooxygenase. Adding a GLA-rich oil that shifts eicosanoid balance in the same direction stacks two antiplatelet influences. Worth stating plainly on a page where the oil is already paired with fish oil, garlic and ginkgo.
Calcium is one of the better-studied single nutrients for comfort across the menstrual cycle and is routinely formulated alongside evening primrose oil. The two mechanisms are unrelated, which is the point of combining them. The calcium half carries the stronger human evidence of the pair.
Chasteberry acts on the prolactin and dopaminergic side while a GLA-rich oil acts on eicosanoid balance, so the two are combined for different reasons in the same formula. This row records the pairing and its rationale. No trial has tested the combination.

Evening primrose oil is pressed from the seed of Oenothera biennis. Roughly seven to ten percent of its fatty acids are gamma-linolenic acid and most of the rest is linoleic acid, so the oil is largely an omega-6 oil with a small distinctive fraction.
GLA skips the pathway's bottleneck. Converting linoleic acid to GLA needs delta-6 desaturase, the slow, tightly regulated step, and supplying GLA directly starts one reaction past it.
GLA is elongated to dihomo-gamma-linolenic acid, and DGLA is the substrate for the series-1 prostaglandins and for 15-HETrE. That is the mechanistic basis for interest in GLA in normal inflammatory and eicosanoid signalling.
DGLA sits at a fork. Delta-5 desaturase can carry it onward to arachidonic acid, which feeds a different eicosanoid series, so where GLA ends up depends on how active that enzyme is.
Where Evening Primrose Oil (PMS) comes from.
Tiny evening primrose seeds are pressed, or washed with a solvent, to get their oil out. It is filtered, sometimes cleaned further until it is pale and odourless, and tested for how much GLA it contains and how fresh it is. Because this kind of oil goes off easily, vitamin E is added and the capsules are filled with air kept out.
Made from a plant. What ends up in the capsule tracks the harvest, so batch testing and a stated marker matter more here than with a made molecule.
The oil comes only from the seed, which is small and low in oil compared with commodity oilseeds. That, plus harvest timing on a plant that sheds seed readily, is why the raw material costs what it does. Growing region and cultivar shift the GLA share within a narrow band.
Three commercial routes. Pressing keeps temperatures low and leaves oil in the meal. Hexane recovers more of it and is then refined. Supercritical CO2 leaves no organic solvent behind at a higher processing cost. Each yields a triglyceride oil with essentially the same fatty acid backbone and a different minor-constituent profile.
All routes filter out seed solids. Refined grades additionally remove phospholipids, pigments and volatile odour compounds, which makes the oil pale and neutral and also strips part of its native tocopherol. Unrefined grades skip this and stay darker.
Batches are assayed by GC and specified on GLA percentage, typically around 9 to 10 percent, with peroxide and anisidine values recorded as oxidation measures. A GLA figure without an oxidation figure describes only half of what matters in a polyunsaturated oil.
The oil is filled into softgels under nitrogen with tocopherol added as an antioxidant, or emulsified and spray dried into a powder for dry formats. Both steps are about keeping oxygen away from a fragile oil.
Labels usually state milligrams of oil without the GLA percentage, which leaves the actual active dose unstated, and they rarely give an oxidation figure or say which extraction route was used.
