Selenium is required for the selenoprotein glutathione peroxidase family, which is highly expressed in testicular tissue and in sperm. The cofactor relationship is established; the pairing effect is not measured.

Pyridoxal 5-phosphate is a cofactor across amino acid and steroid-related metabolism and modulates steroid receptor activity in laboratory work. It is a routine inclusion in male hormonal-support formulas alongside saponin extracts. High-dose pyridoxine over long periods carries a peripheral nerve caution. The cofactor role is settled; the combination effect is not.

DIM influences the balance of hepatic oestrogen hydroxylation pathways through cytochrome P450 induction. Formulas built around fenugreek saponin extracts add it to shift that balance rather than to add anything to the extract's own action. The two act at different points on the hormonal axis. No combination trial is cited.

Calcium D-glucarate inhibits beta-glucuronidase in the gut, which reduces the deconjugation and reabsorption of glucuronidated steroid metabolites. It appears in the same category of formula as Testofen-type extracts for that reason. The mechanism concerns clearance rather than production. Evidence in people is early.

L-citrulline is converted to arginine and raises substrate availability for nitric oxide synthase, which affects blood flow during exercise. Fenugreek saponin extracts are formulated in the same training products but act on a different axis. The combination is additive by formulation intent. No shared pathway links them.

A saponin-standardised extract taken with a calcium dose can reduce the fraction of calcium absorbed at that meal. Spacing the doses is the ordinary formulation answer. The effect is on absorption at that meal, not on mineral status generally.

Fenugreek (Testofen) + Vitamin DEstablished pharmacology

Saponins bind sterols in the gut lumen, and the fat-soluble vitamins depend on intact micelle formation for absorption. Taking a saponin-rich extract with a fat-soluble vitamin at the same meal may reduce the fraction absorbed. Separating them by a meal removes the question.
