Guggulsterones E and Z are geometric isomers of a pregnadiene-dione steroid, and commercial guggulsterone extracts are assayed for the two together.

The starting material is the hardened stem exudate tapped from Commiphora wightii, also catalogued as Commiphora mukul, in the arid regions of north-western India and neighbouring dry zones.
The raw exudate is cleaned of bark, sand and plant debris, in traditional practice by boiling in a triphala or milk decoction and straining, which also removes part of the water-soluble gum.
Ethanol, ethyl acetate or supercritical carbon dioxide pulls the alcohol-soluble resin fraction carrying guggulsterones E and Z away from the gum polysaccharide.
The extract is filtered, the solvent stripped under vacuum, and the concentrate further enriched to raise the guggulsterone share.
High performance liquid chromatography quantifies guggulsterone E and Z, and the concentrate is blended with a carrier until it meets the declared combined percentage on the label.
