The enzyme arginase splits arginine into ornithine and urea, and that ornithine re-enters the same cycle, so the two sit one step apart on the pathway that clears ammonia, a normal byproduct of protein metabolism. Supplying both feeds opposite ends of the loop that keeps nitrogen turnover moving.
Ornithine is the direct substrate that combines with carbamoyl phosphate to form citrulline inside the urea cycle, and citrulline is then recycled back toward arginine and ornithine. Pairing them supports the continuous turning of the cycle the body uses to package and dispose of ammonia.
Arginase cleaves arginine into urea and ornithine, so the two amino acids sit on either side of a single enzyme. Formulators pair them because ornithine regenerates the urea cycle intermediates that arginine turnover consumes.
Ornithine transcarbamylase joins ornithine with carbamoyl phosphate to form citrulline, and citrulline is later recycled back toward ornithine through arginine. The pair covers two adjacent points of the same cycle that handles ammonia nitrogen.
Ornithine is decarboxylated to putrescine, the committed first step of polyamine synthesis, and putrescine is then built into spermidine. Ornithine is the amino acid the body draws on to make this polyamine.
Glutamine is converted to glutamate and then through pyrroline-5-carboxylate to ornithine by ornithine aminotransferase, so it feeds the ornithine pool from upstream. In the gut wall this route is the main endogenous source.
Arginase, the enzyme that splits arginine into ornithine and urea, is a manganese metalloenzyme. Manganese status therefore sets how readily arginine is converted into ornithine.
Pyridoxal 5-phosphate is the cofactor for ornithine aminotransferase, which makes ornithine from glutamate semialdehyde, and for ornithine decarboxylase, which converts it onward to putrescine. B6 status governs traffic into and out of the ornithine pool.
Agmatine is decarboxylated arginine and is hydrolysed by agmatinase to putrescine, the same polyamine that ornithine decarboxylase produces from ornithine. The two enter polyamine synthesis by parallel entry points.
Ornithine and aspartate act at two different steps of the same nitrogen disposal loop. Ornithine accepts a carbamoyl group to become citrulline, and aspartate donates the second nitrogen when citrulline is carried onward. The two are commonly supplied as a single salt for that reason. A 2026 systematic review of the paired form described the human evidence as limited rather than settled, so the pairing rests on biochemistry more than on trial results.
Alpha-ketoglutarate is the amino group acceptor that ornithine aminotransferase works against, so the two intersect directly in glutamate handling. They are sold as a combined salt in sports and clinical nutrition. A 2026 systematic review in adults recovering from burn injury reported mixed metabolic and immune findings for alpha-ketoglutarate, which is where the human record for the pairing sits.
Ornithine aminotransferase converts ornithine to glutamate semialdehyde, which cyclises and is reduced to proline. Ornithine is therefore one of the recognised routes to proline when dietary proline is scarce. Supplying both feeds the same pathway from two directions.
Glutamate and ornithine interconvert through glutamate semialdehyde, with ornithine aminotransferase running the step in either direction depending on tissue and nitrogen load. Glutamate is also the nitrogen donor that feeds carbamoyl phosphate synthesis upstream of ornithine. The relationship is metabolic, not an outcome claim.
Putrescine made from ornithine cannot become spermidine on its own. It needs an aminopropyl group from decarboxylated S-adenosylmethionine, which traces back to methionine. So polyamine synthesis draws on an ornithine arm and a methionine arm at the same time, and either one can be the limiting side.
S-adenosylmethionine is the aminopropyl donor for spermidine and spermine synthesis from putrescine, the polyamine that ornithine decarboxylase produces from ornithine. The two therefore sit on the same committed pathway. This is a pathway relationship rather than a tested combination.
Ornithine, arginine and lysine are all carried by the cationic amino acid transporters of the y+ system, so a large dose of one can occupy uptake capacity the others use. Taking them in the same mouthful means they compete at the gut wall and at cell membranes. Spacing them apart is the usual practical answer.
The steps that surround ornithine in the urea cycle consume ATP, and biological ATP works as a magnesium complex. Magnesium status therefore sits underneath the whole cycle rather than acting on ornithine itself. This is general biochemistry, not a claim about a measured combination.
Zinc availability has been linked in the hepatology literature to the activity of ornithine transcarbamylase, the enzyme that uses ornithine directly. Where zinc intake is low, that link is the reason ornithine has been paired with it. The relationship is described at the enzyme level, not demonstrated as an outcome in a supplementation trial.
Proline made from ornithine only becomes structural collagen after prolyl hydroxylase acts on it, and that enzyme needs ascorbate to keep its iron centre reduced. So ornithine supplies the carbon skeleton and vitamin C permits the modification. Ornithine to proline to hydroxyproline is a documented route; the connective tissue benefit is inferred from it rather than measured for the pair.
Collagen is roughly one third glycine with proline and hydroxyproline making up much of the rest, so glycine and an ornithine derived proline supply feed the same triple helix. Glycine is also the acceptor in creatine synthesis, the reaction that releases ornithine. The pairing is substrate logic; no combination trial supports it.
The first step of creatine synthesis transfers the guanidino group from arginine to glycine, and the leftover carbon skeleton is ornithine. Taking creatine reduces the pull on that endogenous route, which changes how much ornithine the body generates that way. The connection is directional biochemistry rather than a performance claim.
A large protein load raises the amount of amino nitrogen that has to be converted to urea, and ornithine is the carrier that cycle turns on. Pairing the two is common in sports nutrition for that reason. It is a plausible metabolic fit, not an effect that has been measured for the combination.
Gut bacteria both generate ammonia from urea and produce polyamines from ornithine, so the microbial community sits between an oral ornithine dose and what reaches the liver. Changing that community with live cultures could plausibly change the arithmetic. The evidence here is mechanistic and early.
