- Start with 200 to 500mg a day of root extract, taken with a meal that contains fat since the diterpenes need bile to be picked up. 1,000mg is a trial condition, not a daily target.

The requirement for a lipid phase is established; the size of any exposure difference for this extract is not quantified here.
Phospholipids emulsify lipophilic extracts, keeping them dispersed in a formula and supporting micelle formation during digestion. Coleus extracts are resinous and settle out of aqueous formats without an emulsifier. This is a delivery role and adds nothing pharmacologically of its own.
Coleus and rosemary are both Lamiaceae and both carry rosmarinic acid, a caffeic acid ester that is a major constituent of non-forskolin Coleus preparations. Combining them raises the total rosmarinic acid in a formula from two sources. Total intake should be counted across both rather than per ingredient.
Coleus preparations are usually formulated into stimulant-containing blends, and L-theanine is the conventional counterweight added to smooth the subjective feel of those blends. The relationship is with the stimulant in the formula, not with Coleus itself. This is formulation convention rather than a measured interaction.
Both ingredients contribute polyphenolic constituents to a formula, with different structural classes and different absorption behaviour. Neither substitutes for the other, and the combined polyphenol load is what a label should reflect. The basis is compositional overlap rather than a tested pairing.

Coleus barbatus is a Lamiaceae species whose roots accumulate labdane-type diterpenes, built through the plant terpenoid pathway from geranylgeranyl diphosphate.
A non-forskolin preparation is defined by what it excludes: it is standardised on other root constituents rather than on forskolin, so the adenylyl cyclase pharmacology associated with forskolin is not the basis of the ingredient.
Labdane diterpenes are strongly lipophilic and depend on bile-driven micelle formation with dietary fat for intestinal uptake, which is why these extracts are usually presented in oil or with an emulsifier.
Because the diterpene and phenolic fractions partition differently, the solvent used sets the constituent profile: a hydroethanolic extract and a supercritical carbon dioxide extract of the same root are not interchangeable materials.
