In a four week, three arm randomised, double-blind, placebo-controlled trial, 128 adults self-reporting low mood but not diagnosed with depression took a standardised saffron stigma extract (affron) at 28 mg per day, 22 mg per day, or placebo, with POMS, PANAS and DASS-21 measured at baseline and end. Negative mood and symptoms related to stress and anxiety fell at 28 mg per day, with a difference against placebo on the POMS Total Mood Disturbance scale (p < 0.001, d = -1.10), and no treatment effect at 22 mg per day. Screening and outcomes were self-reported, which the authors name as the main weakness.

Mood formulas often combine them for that reason, and because both lean the serotonin system the same way it is sensible not to stack many serotonin-supporting ingredients at once.

Applied to saffron the reasoning is inferred from piperine's general pharmacology rather than measured on crocetin. Because piperine broadens exposure to anything sharing those routes, its presence is a reason to review other things a person takes.

Saffron is the dried stigma of Crocus sativus. Its characteristic constituents are the crocins, water-soluble carotenoid glycosides of crocetin, plus picrocrocin and its aglycone safranal, which carries the aroma.
Crocins are hydrolysed in the gut lumen and at the enterocyte to crocetin, which is the lipophilic form that enters circulation; measured saffron exposure in blood is therefore largely crocetin and its conjugates rather than intact crocin.
Crocetin is a dicarboxylic carotenoid, so unlike lutein or beta-carotene it carries charged end groups and behaves partly like a water-soluble carotenoid, which is why saffron extracts disperse readily in aqueous systems.
Safranal and crocetin are both electron-rich conjugated systems and quench singlet oxygen and peroxyl radicals in lipid phases, the ordinary chemistry of a polyene carotenoid.
Where Saffron Safr-Inside comes from.
It is the red threads from a crocus flower, picked and dried by hand, then soaked in a water and alcohol mix to pull out the colour and aroma compounds. The extract is tested by lab instrument so each batch carries the same amount of the compounds the label names. Saffron is costly for one plain reason: it takes an enormous number of flowers to make a small amount of it.
