Diallyl disulfide does not exist in the intact clove. Crushing garlic lets alliinase act on alliin to form allicin, which decomposes to diallyl disulfide and related sulfides, so garlic preparation determines how much DADS a product can contain. Pairing the isolate with garlic material is a source relationship, not two independent actives.
Ageing garlic in aqueous ethanol shifts the sulfur profile toward water-soluble species such as S-allylcysteine and away from the lipid-soluble sulfides, so aged extract is low in diallyl disulfide by design. Combining the two gives a wider sulfur spread rather than more of one compound. The clinical consequence of that spread has not been isolated.
Diallyl disulfide is reduced by glutathione and thiol groups, and that reaction is what releases its hydrogen-sulfide-donating and allyl-thiol species. N-acetylcysteine supplies cysteine for glutathione synthesis, so thiol supply sits upstream of DADS activation. Because the same reaction consumes reduced thiols, the two are coupled in both directions.
Glutathione is the direct reductant that opens the disulfide bond in diallyl disulfide, generating allyl perthiol and hydrogen sulfide. Without a pool of reduced glutathione the compound stays largely unreacted. This is a mechanistic dependency described in cell systems rather than a benefit shown in people.
Both compounds are electrophilic and both act on the Keap1-Nrf2 axis to raise phase II enzyme expression, including glutathione S-transferases and NAD(P)H quinone oxidoreductase. Stacking them means two inputs to one transcriptional response, which can be additive but can also plateau. The reported work is largely cell and animal, so this is mechanism rather than a human outcome.
Curcumin is another electrophile that engages Nrf2-dependent antioxidant enzyme expression and dampens NF-kB signalling in cell models. The overlap with diallyl disulfide is at the same nodes rather than at complementary ones. No combination trial in people supports the pairing.
The thiol chemistry that diallyl disulfide sets in motion feeds into glutathione peroxidase activity, and every glutathione peroxidase is a selenoenzyme. Adequate selenium status is therefore a condition for that arm of the response to run. Garlic grown in selenium-rich soil also accumulates selenium compounds, which is a separate reason the two travel together.
Alpha-lipoic acid cycles between its dithiol and disulfide forms and helps regenerate reduced glutathione, the same pool that diallyl disulfide draws on when it reacts. The pairing supports thiol turnover rather than adding a second independent action. Measured in cell and animal systems, not in a human combination trial.
Garlic sulfides and long-chain omega-3 fatty acids both reduce platelet aggregation in laboratory measures, by different routes. Taken together those laboratory effects point the same way, which is worth flagging for anyone already on an antiplatelet or anticoagulant medicine or scheduled for a procedure. This is an additive caution, not a benefit claim.
Ginger constituents inhibit thromboxane-mediated platelet aggregation in laboratory assays, the same functional endpoint garlic sulfides move. Stacking the two culinary botanicals at supplemental doses points in the same direction. The clinical size of the combined shift has not been quantified.
Nattokinase acts on fibrin degradation while garlic sulfides act on platelets, so the two touch different parts of haemostasis in the same direction. Flag the combination rather than assume it is neutral. No trial of the pair is cited.
MSM is a stable oxidised sulfone that contributes sulfur without the reactive disulfide chemistry diallyl disulfide brings. The two are combined in sulfur-themed joint and skin formulas. Nothing in the cited evidence measured them together, so this is formulation rationale.
Boswellic acids and garlic sulfides appear together in blends aimed at joint comfort and mobility during activity. The candidate garlic literature on joint tissue is animal work and a narrative review, so the pairing rests on formulation habit rather than a measured combined effect in people.
Quercetin is abundant in onion and garlic tissue and, like diallyl disulfide, modulates NF-kB-linked signalling in cell models. Whole-allium intake therefore delivers both, which is one reason food-level effects are hard to attribute to a single sulfide. Isolated-compound combination data in people is absent.
High-dose alpha-tocopherol has its own reported antiplatelet effect and also sits in the lipid compartment where oil-soluble garlic sulfides distribute. Combining them adds two mild influences in the same direction. Worth noting before a procedure rather than a reason to pair them.

Where Diallyl Disulfide DADS comes from.

The same molecule is reached more than one way. Which route a given product used is a manufacturing choice, and the finished compound is the same either way.
Intact cloves carry alliin and the enzyme alliinase in separate compartments, so the bulb itself contains essentially no diallyl disulfide.
Mechanical damage lets alliinase act on alliin to form allicin, the immediate precursor.
Allicin is unstable and breaks down to diallyl disulfide, diallyl trisulfide, ajoene and vinyldithiins, with heat and time deciding the mix.
