Whole garlic is where allicin comes from: the bulb stores alliin and keeps alliinase in a separate compartment, and the two only meet when tissue is crushed. A formula pairing a stabilised allicin with garlic powder is stacking a finished thiosulfinate on top of a substrate-plus-enzyme system. The two deliver sulfur chemistry on different timelines, which is worth stating rather than adding the numbers together.

Ageing garlic in aqueous ethanol converts the reactive thiosulfinates into stable water-soluble compounds, chiefly S-allylcysteine, so aged extract carries almost no allicin. Combining the two gives a broader sulfur profile rather than a higher allicin dose. Neither substitutes for the other on an assay.

Allicin reacts fast with free thiols by thiol-disulfide exchange, and glutathione is the most abundant free thiol in a cell. The reaction forms S-allylmercaptoglutathione and consumes the allicin in the process. Co-dosing a large free-thiol load in the same capsule therefore works against delivering intact allicin, which is a formulation consideration and not a safety warning.

Dietary nitrate is reduced to nitrite and then nitric oxide by oral bacteria and tissue enzymes. Allicin is broadly antimicrobial, so a high allicin dose taken at the same time could in principle interfere with the oral nitrate-reducing flora that beetroot depends on. That gives this pair two directions at once, and neither has been measured in a co-dosing trial.

Both are used in formulas aimed at supporting normal vascular function, and each has its own literature on blood pressure measures. The mechanisms are unrelated, so any combined effect would be simple addition. No study has looked at the pair.

Red yeast rice acts on hepatic cholesterol synthesis; garlic sulfur compounds have been studied for effects on circulating lipid measures by other routes. Allicin is not stored in garlic. Alliin sits in the cytosol and the enzyme alliinase sits in the vacuole, and only when tissue is crushed do they meet, at which point allicin forms within seconds.

Allicin is a thiosulfinate and chemically unstable. It rearranges into diallyl disulfide, diallyl trisulfide, ajoene and vinyldithiins, so a finished material's sulfur profile reflects how it was handled after formation as much as the garlic it came from.

Allicin reacts with free sulfhydryl groups by thiol-disulfide exchange, transferring an allyl-thio group to cysteine or glutathione. Most of its cellular chemistry follows from that single reaction type.
