Glycerol is a three-carbon sugar alcohol, propane-1,2,3-triol, fully miscible with water and strongly hygroscopic.
Once absorbed, glycerol distributes through total body water and is osmotically active, so plasma osmolality rises and renal free water clearance falls for a period after ingestion. That transient reduction in urine output is the basis for its use in pre-exercise fluid loading.
Glycerol crosses membranes both by simple diffusion and through the aquaglyceroporin channels AQP3, AQP7 and AQP9, which is how adipose tissue exports it and the liver takes it up.
Hepatic glycerol kinase phosphorylates glycerol to glycerol-3-phosphate, which either enters glycolysis and gluconeogenesis by way of dihydroxyacetone phosphate or is re-esterified with fatty acids into triglyceride.
Where Glycerol (HydroMax) comes from.
Glycerol is the backbone left over when a fat or oil is split apart, which happens on a large scale when biodiesel or soap is made. The raw stream is distilled and filtered until it meets a pharmacy-grade specification, including a test for the contaminant that grade exists to catch. To turn the syrup into a powder, it is sprayed onto a fine carrier until it pours.
The same molecule is reached more than one way. Which route a given product used is a manufacturing choice, and the finished compound is the same either way.
Palm, coconut, soybean or rapeseed oil are the usual plant sources; tallow is the animal one. The feedstock determines whether the finished glycerol can be declared vegetable or kosher or halal, and it is the reason a certificate of origin is asked for.
Three routes free the glycerol from the fat: transesterification with methanol during biodiesel manufacture, which leaves crude glycerol as the co-product; high-pressure hydrolysis with water in fat splitting; and saponification with alkali in soap making. A separate petrochemical route from propylene exists and a fermentation route from sugar is used at smaller scale.
Crude glycerol carries salts, methanol residues, free fatty acids and colour. Vacuum distillation, ion exchange and activated carbon bleaching bring it to the USP or pharmaceutical grade with the specification on residual methanol and on diethylene glycol.
Finished liquid is assayed for glycerol content, water, ash, heavy metals and specifically for diethylene glycol, the contaminant that pharmacopoeial glycerol testing exists to exclude.
To make the powder, the purified liquid is sprayed onto and blended with silicon dioxide or maltodextrin until it flows freely, then sized and packed under low humidity. The declared glycerol percentage is set at this step.
