Vitamin B12 is made by bacteria and archaea, not by plants, so a diet with no animal foods carries no reliable source of it. Supplemental or fortified B12 is the standard way the intake is met. This is settled nutrition biochemistry and does not rest on a single trial.
Plant diets supply alpha-linolenic acid, and the enzymatic conversion of alpha-linolenic acid to DHA in humans is limited. Algal oil supplies DHA already formed, bypassing the conversion step. Plasma and red cell DHA are markers of status, not outcomes in themselves.
Conversion of alpha-linolenic acid onward to EPA proceeds further than conversion to DHA but is still constrained by desaturase and elongase capacity. Algal oils are formulated to supply EPA alongside DHA for that reason. The rationale is metabolic, and the measure is a blood fatty acid level rather than a clinical endpoint.
Cholecalciferol is conventionally made from lanolin, and lichen-derived cholecalciferol is the plant-source equivalent used in vegan formulas. Both raise serum 25-hydroxyvitamin D, which is the accepted status marker. Sun exposure is the other input and it varies with latitude and season.
Ascorbate reduces ferric iron to the ferrous form and holds it soluble through the small intestine, which raises fractional absorption of non-heme iron from plant foods. Plant diets carry only non-heme iron, so the enhancer matters more here than on a mixed diet. Timing with the iron-containing meal is what makes it work.
All iron in a plant diet is non-heme iron, whose absorption is far more sensitive to meal composition than heme iron is. Supplemental iron in a vegan stack is usually included on that basis rather than because intake is low. Serum ferritin is a status marker and should be read as one.
A substantial calcium dose taken in the same sitting reduces absorption of non-heme iron and of zinc. Vegan stacks tend to carry both minerals, which puts them in the same capsule at the same time. Splitting calcium away from the iron-containing dose is the usual formulation answer.
Phytate in legumes, grains and seeds binds zinc in the gut lumen and lowers the fraction absorbed, and the phytate to zinc molar ratio of plant diets is high. Supplemental zinc is included in vegan formulas against that background. Soaking, sprouting and fermenting food lowers phytate and moves the same lever.
Phytase hydrolyses inositol hexaphosphate and releases the minerals it had chelated, which is why the enzyme is added to plant-heavy meals and to some formulations. The effect is on mineral availability from the meal, not on the supplement's own mineral salts. It has to be present in the gut at the same time as the phytate.
Iodine content of plant foods tracks soil iodine and is unreliable, and the usual dietary anchors are dairy, fish and iodised salt. Sea vegetables carry iodine but at concentrations that swing by orders of magnitude between batches. Supplemental potassium iodide gives a defined amount, which is the reason it appears in vegan stacks.
Selenium enters the food chain from soil, so plant selenium content varies enormously by growing region. The element is required for glutathione peroxidase and the deiodinase enzymes. Brazil nuts are the common plant source and they are the least consistent one.
The densest dietary choline sources are eggs and liver, and plant sources are more dilute, so intake tends to sit lower without them. Choline is needed for phosphatidylcholine and acetylcholine synthesis and for hepatic fat export. Endogenous synthesis exists but does not cover the requirement.
Dietary creatine comes from meat and fish, so intake on a plant diet is close to zero and the body relies on endogenous synthesis from glycine, arginine and methionine. Muscle creatine content is measurably lower in vegetarians than in omnivores, which is a status marker. Supplementation raises the muscle pool.
Taurine in the diet comes almost entirely from animal tissue and shellfish, and humans make it from cysteine by way of cysteine sulfinic acid decarboxylase. Plasma and urinary taurine sit lower on plant diets, which is an association with intake rather than a demonstrated consequence. Supplemental taurine restores the intake side.
Red meat is the dominant dietary carnitine source, and the body synthesises carnitine from lysine and methionine using iron, vitamin C and niacin as cofactors. Plasma carnitine tends to be lower in long-term vegetarians, a marker of intake. Carnitine carries long-chain fatty acids into the mitochondrion.
Muscle carnosine is made from beta-alanine and histidine, and beta-alanine in the diet comes from meat and fish. Vegetarian muscle carnosine content measures lower, which is a tissue marker. Supplemental beta-alanine supplies the rate-limiting precursor directly.
Dairy and eggs are principal riboflavin contributors in typical diets, so removing them lowers one of the main inputs. Riboflavin is the precursor of FAD and FMN, cofactors in energy metabolism and in the enzyme that recycles folate. Fortified foods and supplements are the substitute source.
Plant diets are generally high in folate, and high folate intake can normalise a blood count while B12 status stays low, which is why B12 must be assessed on its own markers. Methylmalonic acid and holotranscobalamin are the more specific indicators. Both nutrients feed the same one-carbon cycle, so folate cannot substitute for B12 in the methionine synthase step.
MK-7 is produced by Bacillus subtilis natto fermentation, which makes it one of the few menaquinone sources that is not animal-derived. Menaquinones carboxylate osteocalcin and matrix Gla protein, so they act on how the body incorporates calcium into bone tissue. Plant diets supply phylloquinone readily but long-chain menaquinones far less so.
Lysine is the limiting amino acid in cereal grains, so a plant diet built mainly on grains reaches its lysine ceiling before its total protein ceiling. Legumes are lysine-rich, which is the classical reason grains and legumes are combined. Lysine is also a precursor for carnitine synthesis.
Plant protein isolates generally carry less leucine per gram than whey does, and leucine is the amino acid that triggers the muscle protein synthesis signal. Formulators add free leucine to plant protein blends to close that compositional gap. This is about amino acid composition, not protein quantity.
Tannins bind non-heme iron in the gut lumen and form complexes that are not absorbed, and tea and coffee taken with a meal are the everyday version of this. A plant diet, which carries only non-heme iron, is more exposed to the effect. Separating tea from the iron-containing meal is the practical response.
Catechins chelate non-heme iron in the intestinal lumen, lowering the fraction absorbed from the same meal. The interaction is with dietary and supplemental non-heme iron, not with body iron stores. Spacing the extract from the iron dose is what formulators do about it.
