Rutin and its hydroxyethylrutoside derivatives have a long pharmacology literature on capillary resistance measures. They pair with centella on the same complementary logic as diosmin. What is measured in most of that work is a marker such as capillary filtration, not a symptom endpoint.
Arginine is the substrate endothelial nitric oxide synthase uses to make nitric oxide, the main endothelium-derived vasodilator. Citrulline escapes the intestinal and hepatic arginase that clears much of an oral arginine dose, then is converted to arginine in the kidney, which is why it raises plasma arginine more reliably per gram. Proline and its hydroxylated form make up roughly a fifth of the residues in collagen, so proline supply is a substrate input to collagen synthesis. Gotu kola triterpenes act on the transcription and secretion side in fibroblasts. Substrate and signal are different levers on the same process.
Every third residue in the collagen triple helix is glycine, because only glycine is small enough to sit at the helix core. That makes glycine the single most abundant amino acid in collagen by count. It is a substrate contribution, not a signal, and it belongs alongside the triterpene rather than duplicating it.
Magnesium acts as a natural calcium antagonist at vascular smooth muscle, competing with calcium at channel and binding sites, which is why magnesium status affects vascular tone. Centella asiatica carries pentacyclic triterpenes as its characteristic constituents: the glycosides asiaticoside and madecassoside, and their aglycones asiatic acid and madecassic acid.
Asiaticoside and madecassoside are glycosides that must be hydrolysed, partly by gut microbial glycosidases, to their aglycones before systemic exposure, so the circulating profile differs from the label profile.
Collagen maturation requires prolyl and lysyl hydroxylases, which are ascorbate-dependent iron-containing enzymes, so any signal that raises collagen transcription still depends on adequate vitamin C and iron to produce a stable helix.

The leaves and stems of a low creeping plant are dried and ground, then soaked in a mix of alcohol and water that pulls out the active compounds. That liquid is filtered and concentrated down, and the concentrate is tested so the label can state how much of the active triterpenes it holds. It then goes into capsules, tablets or a dropper bottle.
Made from a plant. What ends up in the capsule tracks the harvest, so batch testing and a stated marker matter more here than with a made molecule.
Leaf and stem harvested from cultivated or wild-collected plants, chiefly in South and Southeast Asia and Madagascar; the plant grows in wet ground, which makes source-water and heavy metal screening relevant.
