Glycyrrhizin is the licorice constituent that inhibits renal 11-beta-HSD2, so cortisol acts on the mineralocorticoid receptor and the kidney trades potassium out for sodium. A glycyrrhizin-standardized extract carries this at full strength, unlike a deglycyrrhizinated one, so potassium status moves in a predictable direction.
The mineralocorticoid activity glycyrrhizin unmasks increases renal excretion of magnesium alongside potassium, since both follow the same sodium-retaining, cation-wasting pattern in the distal tubule. Magnesium status is worth tracking when a glycyrrhizin-standardized extract is taken over weeks rather than days.
Marshmallow root mucilage coats the mucosal surface while the licorice fraction acts on the mucus the lining secretes itself. Glycyrrhizin-standardized material is normally dosed lower in these blends than deglycyrrhizinated licorice, for the mineralocorticoid reason above.
Glycyrrhizin's active metabolite inhibits 11-beta-hydroxysteroid dehydrogenase type 2, so cortisol acts at the mineralocorticoid receptor and the kidney sheds more potassium and magnesium. Magnesium intake offsets that loss.
The same receptor effect promotes sodium and water retention. Added sodium pushes normal fluid balance the same way, and a standardised glycyrrhizin extract makes this stronger than whole root.

Glycyrrhizin is a triterpenoid saponin: the aglycone 18-beta-glycyrrhetinic acid carrying a disaccharide of two glucuronic acid units.
Glycyrrhizin taken by mouth is largely not absorbed intact; gut bacterial beta-glucuronidase strips the sugars and the released 18-beta-glycyrrhetinic acid is the absorbed active species.
18-beta-glycyrrhetinic acid inhibits 11-beta-hydroxysteroid dehydrogenase type 2, the enzyme that normally converts cortisol to inactive cortisone in the kidney, so cortisol occupies the mineralocorticoid receptor and sodium retention with increased potassium excretion follows. This is why intake ceilings for glycyrrhizin exist in food and monograph guidance.
Glycyrrhizin is roughly fifty times sweeter than sucrose by weight with a slow onset and long finish, which is why licorice extract functions as a sweetener and flavour modifier as well as an active.
Where Licorice Root (Glycyrrhizin) comes from.
The root is dug up after a few years, washed, dried and simmered or soaked to pull the actives out. That liquid is concentrated and tested for glycyrrhizin, the sweet compound. If the maker wants DGL, an extra step strips that compound out before drying; if not, the extract is dried as is.
Made from a plant. What ends up in the capsule tracks the harvest, so batch testing and a stated marker matter more here than with a made molecule.
Roots and underground runners of Glycyrrhiza glabra, Glycyrrhiza uralensis or Glycyrrhiza inflata, usually lifted after three to four growing seasons. Species and growing region both move the glycyrrhizin level in the raw material, which is why assay comes before formulation.
Roots are washed, cut and dried to a stable moisture level. Metal uptake from the soil is a raw-material property of this crop, so incoming testing on heavy metals is part of normal receiving.
Water pulls the saponins and polysaccharides; adding ethanol brings across more of the flavonoid fraction including glabridin. The solvent choice sets which constituents dominate the finished extract.
For deglycyrrhizinated material the extract is treated so glycyrrhizin partitions out, commonly by acidification and solvent partition or by ammonia treatment and filtration. Residual glycyrrhizin is then assayed against the declared limit.
The concentrate is assayed by HPLC for glycyrrhizin, and for DGL against the residual limit, then adjusted with carrier to the declared percentage.
Spray drying or vacuum drying onto a carrier such as maltodextrin or gum arabic gives the free-flowing powder that goes into capsules and tablets. Block or paste extracts are the older confectionery-facing format of the same stream.
