- Suits people who want the named glucan fraction with a beta-glucan figure on the label. Dried shiitake carries the same polysaccharide if you would rather get it from food.

- Start with 100mg to 500mg a day of extract standardised to beta-glucan. Read the beta-glucan number, since a plain polysaccharide figure can be largely substrate starch.

Ascorbate accumulates in leukocytes at high concentration and supports normal neutrophil chemotaxis and oxidative burst control. It works in the aqueous phase while a beta-glucan works at the receptor level, so the two do not overlap. Established nutrient biochemistry, not a tested pair.
Elderberry anthocyanins and fungal beta-glucans sit in the same category of product for different reasons. The pairing is commercial and traditional rather than mechanistically linked. Nothing has measured the two together.
Propolis contributes flavonoids and phenolic acids, shiitake contributes a polysaccharide. They are combined by convention. Confidence is early and the rationale is compositional breadth.
Much of an oral beta-glucan is not absorbed intact and reaches the colon, where it is fermented. That makes a fungal polysaccharide a plausible substrate alongside a live culture. Fermentability is established; which strains benefit and by how much is not settled.
Inulin and fungal beta-glucans are both fermented in the colon to short-chain fatty acids, though at different rates and by different organisms. Combining substrates broadens which bacteria are fed. The fermentation chemistry is established; a combined effect on any immune measure is not.

Where Shiitake Lentinan comes from.
The mushroom is grown, dried and simmered in hot water, which pulls out the long-chain sugar that lentinan is. Alcohol is then added to make that sugar drop out of solution so it can be collected and dried. The number worth reading on the label is beta-glucan measured on its own, because a plain polysaccharide figure can be mostly starch from the grain the fungus was grown on.
The same molecule is reached more than one way. Which route a given product used is a manufacturing choice, and the finished compound is the same either way.
Fruiting bodies are grown on hardwood logs or supplemented sawdust blocks. Mycelium is grown either on sterilised grain or in submerged liquid culture. These are genuinely different starting materials and the label should say which one was used.
Fruiting bodies are dried before extraction. Ultraviolet exposure during drying converts ergosterol to vitamin D2, which is why some shiitake products carry a vitamin D2 figure and others do not.
Water-soluble beta-glucans are pulled out at elevated temperature. Chitin and other insoluble wall material stays behind. Some processes add an alkaline extraction step to release more tightly bound glucan.
Ethanol is added to the aqueous extract to precipitate high molecular weight polysaccharide while leaving small molecules and sugars in solution. Repeated cycles raise the glucan share.
An enzymatic assay measures beta-glucan specifically and subtracts alpha-glucan starch. This distinction is the whole game on a mushroom label, since a total polysaccharide figure can be mostly starch from a grain substrate.
Dried and blended into capsules, tablets or powders. The oral form is what supplements use; the clinical literature that made lentinan known used an injected preparation under medical supervision, which is a different route entirely.
Whether the material is fruiting body or grain-grown mycelium is frequently not stated, and the two carry different amounts of beta-glucan.
