Pantothenic acid becomes coenzyme A, needed to build the cholesterol the adrenal cortex converts into its steroid hormones. It is the second fixture of these formulas after vitamin C.
Pyridoxal-5-phosphate is the cofactor for aromatic L-amino acid decarboxylase, the step that makes dopamine on the way to adrenal catecholamines. It serves the medullary arm while B5 and vitamin C serve the cortical arm.
Magnesium restrains NMDA receptor activity and is reported to damp ACTH release from the pituitary, the upstream signal that drives cortical output. Urinary magnesium losses also rise while the stress axis is running hot.

Cells with low niacin status have less reducing power to spend on steroid synthesis. The relationship is textbook cofactor biochemistry rather than a tested combination.
Zinc sits in the structural core of the nuclear receptors that steroid hormones signal through, including the glucocorticoid receptor zinc fingers. It is also a cofactor for enzymes across the wider endocrine tissue. This is receptor and enzyme architecture, so it is about the machinery working normally rather than about raising any hormone.
Blends of this type often carry licorice root, whose glycyrrhetinic acid inhibits 11-beta-hydroxysteroid dehydrogenase type 2 in the kidney and leaves cortisol acting at the mineralocorticoid receptor. The predictable consequence is sodium and water retention with potassium loss. Anyone watching sodium intake should regard the two as acting in the same direction.
A dose-response meta-analysis of probiotic supplementation pooled trials that measured circulating cortisol, a marker of hypothalamic-pituitary-adrenal activity rather than an outcome in itself. That places gut-directed supplements on the same axis these blends target. The two have not been trialled together as a single product.
Cortisol and melatonin run on opposed daily rhythms, cortisol rising toward waking and melatonin toward the dark phase. Formulators split them across the day for that reason, adaptogenic blends in the morning and melatonin at night. Taken together at the same hour they pull the same axis in two directions.

Eleuthero was the herb the adaptogen category was originally defined around and appears in most commercial adrenal blends. Its eleutherosides are chemically unrelated to withanolides or rosavins, so the herbs are not redundant. There is no combination trial of the finished blend.
Schisandra lignans are a standard fourth or fifth herb in these formulas and also induce phase II detoxification enzymes in laboratory models. That gives it a role distinct from the cortisol-facing herbs. The pairing is formulation practice.

Adrenal steroid synthesis begins when cholesterol is moved into the mitochondrion and cleaved to pregnenolone by CYP11A1, the rate-limiting step for every downstream corticosteroid.
Pantothenic acid is the backbone of coenzyme A, and coenzyme A is required for the cholesterol supply that feeds steroid synthesis, which is why B5 is standard in these blends.
The adrenal cortex holds one of the highest tissue concentrations of ascorbate in the body, and ascorbate is consumed during steroid hydroxylation, which is the reason vitamin C sits in almost every formula of this type.

Nothing here is made in one place. Each herb is grown, dried and extracted on its own, the vitamins are made separately in a factory, and the finished capsule is those powders mixed together. What is actually in it depends entirely on the individual inputs the blender bought.
The same molecule is reached more than one way. Which route a given product used is a manufacturing choice, and the finished compound is the same either way.
Ashwagandha root, rhodiola root, holy basil leaf and licorice root arrive as dried crude botanicals from separate growers, typically India, China, Eastern Europe or Central Asia depending on the species.
Calcium pantothenate, ascorbic acid and pyridoxine come from established chemical or fermentation routes and enter the blend as purified powders, not from the plants.
Each botanical is extracted separately, most often with water, ethanol or a water-ethanol mix, then the solvent is removed under vacuum.
Extracts are assayed by HPLC against a marker such as withanolides, salidroside or glycyrrhizin, then diluted with carrier or blended with a stronger lot to hit the stated percentage.
