In a balanced crossover trial, 10 healthy men ingested 70, 140 or 280 mL of concentrated beetroot juice, carrying 4.2, 8.4 and 16.8 mmol inorganic nitrate, with resting plasma nitrate and nitrite tracked over 24 hours. Plasma nitrite rose dose dependently and the peak change occurred at approximately 2 to 3 hours after ingestion. Exercise testing was timed to 2.5 hours postingestion on that basis, where 280 mL lowered steady state oxygen uptake during moderate cycling by 3.0 percent, with a non-significant 1.7 percent reduction at 140 mL.

The two act on different limits during hard effort, with beetroot nitrate supporting blood flow and the oxygen cost of muscle work while beta-alanine raises muscle carnosine that buffers the acid load of intense contractions. Because the mechanisms do not overlap, the two are routinely formulated together.
Quercetin scavenges superoxide, which is the species that inactivates nitric oxide fastest by forming peroxynitrite. Less oxidant pressure means more of the nitrite-derived nitric oxide survives to signal.
Arginine is the direct substrate of nitric oxide synthase, while beet nitrate is reduced to nitrite and then to nitric oxide without an enzyme step. Both converge on the same signalling molecule.

Those bacterial reductases carry a molybdenum cofactor at their active site. That makes molybdenum an enabling part of the chemistry rather than an additive one, and the relationship is enzymology rather than a clinical finding in people taking both.
Nitrate from beetroot does nothing until bacteria reduce it, and the species that do this live on the dorsal tongue rather than in the colon. Most swallowed probiotic strains are selected for the gut, so an oral-cavity strain is the one with a mechanistic rationale here. Read the pairing as mechanistic rather than clinical, since the step being described is microbial conversion and not a measured outcome in people.
Garlic organosulfur compounds influence nitric oxide signalling and hydrogen sulfide chemistry in vascular tissue. Beetroot supplies the substrate for the reduction route to nitric oxide. The overlap is mechanistic; the pairing supports normal vascular tone rather than acting on any named condition.
Bicarbonate works as an extracellular buffer against hydrogen ion accumulation, while beetroot nitrate works on oxygen cost and vascular tone. They do not share a pathway, which is exactly why they are stacked. Gastrointestinal tolerance of bicarbonate is the practical limit on the combination.
Oxalate forms poorly soluble complexes with divalent cations before they reach the absorptive surface. Non-heme iron taken in the same sitting as a large beetroot dose is the cation most often affected in practice. Separating the two by a couple of hours is the standard formulation answer, and this is chemistry rather than a trial result.
Calcium binds oxalate tightly, so the two reduce each other's free availability when taken together. The same reaction is the reason dietary calcium lowers oxalate absorption from the gut. Whether that is wanted or unwanted depends on which of the two the formula is trying to deliver.
Beetroot root is itself a source of naturally occurring folate, which is why it appears in reviews alongside folic acid nutrition. The cited paper is a review, not a controlled comparison, so this is background rather than measured effect. It supports normal folate intake as a food source, and nothing stronger should be read into it.

The part of beetroot that drives blood vessel signalling is inorganic nitrate, not a polyphenol. You absorb it, your salivary glands concentrate it and send it back into your mouth, and bacteria on your tongue turn it into nitrite.
That nitrite is then converted into nitric oxide, a step that happens more readily where oxygen is low and acidity is high. Oxygen-depleted proteins in your blood and muscle drive it, so this route works where the usual enzyme route works least well.

That first conversion step in your mouth is done by bacteria and their enzymes. Antibacterial mouth rinsing wipes those organisms out, and that's the main reason the same nitrate dose behaves so differently between people.

Nitrate is measured by ion chromatography or a colorimetric assay and adjusted with carrier to a declared milligram figure. Products standardised on betalains use a spectrophotometric pigment reading instead, and the two figures are not interchangeable.
