L-theanine is made enzymatically from L-glutamine and ethylamine, or extracted from Camellia sinensis leaf. Caffeine comes from the extract streams of coffee and tea decaffeination, or is synthesised from urea and a xanthine intermediate by the classical purine route.
For theanine, microbial glutaminase couples the ethylamine to glutamine and yields the single L-enantiomer directly, which is what allows an enantiomeric purity specification. Purely chemical synthesis gives a racemic DL mixture requiring resolution. Synthetic caffeine is built up through methylation steps to the trimethylxanthine.
Tea leaf is extracted with hot water or aqueous ethanol, which pulls caffeine, theanine and catechins together. Separating them requires further chromatography; an extract that skips that step delivers all three in the leaf's own ratio.
