In a randomised, open-label crossover pharmacokinetic study, 12 healthy volunteers took single doses of two milk thistle supplement formulations, with blood sampled over 8 hours. Silybin A and B were rapidly absorbed and eliminated in both, and the soft-gel formulation reached higher peak and total exposure than the powdered capsule.

What Milk Thistle actually does.

Milk thistle seed gives you silymarin, which isn't one molecule but a mix of flavonolignans: silybin A and B, isosilybin, silychristin, silydianin, plus the flavonoid taxifolin. The proportions differ between extracts, so two products declaring the same silymarin percentage can still differ in composition.

Silybin is the constituent studied most often and usually makes up the largest share of silymarin, which is why some extracts are standardised to silybin as well as to total silymarin.

Silymarin dissolves poorly in water and gets heavily conjugated by glucuronidation and sulfation on first pass, so blood levels after an oral dose stay low and don't last long. That limit is the reason phospholipid complexes and lipid vehicles exist.

In laboratory systems, flavonolignans donate hydrogen atoms to lipid radicals, which is the chemical basis for calling them chain-breaking antioxidants. Antioxidant capacity measured in a test tube is a chemical property, not a demonstration of an effect in your tissue.

The seeds are cleaned and their oil pressed out, then the active compounds are pulled from the leftover meal with a solvent. The liquid is boiled down and the solids recovered, measured for how much silymarin they contain, and finished as a powder or worked into a fat or phospholipid base to help them dissolve.
